What is it? / Overview
Guggulsterones are proposed to upregulate hepatic LDL-receptor activity and bile-acid excretion, and directly stimulate the thyroid in animal studies -- but a rigorous American RCT found no cholesterol benefit (and possibly an LDL increase), in contrast to older, more positive studies. The thyroid-support marketing claim rests on animal data, not human trials.

The CYP3A4-induction interaction potential is broad enough to be worth a pharmacist check for anyone on regular medications -- this is one of the more interaction-heavy botanicals in this database.

Who should avoid it?
Avoid with: Breastfeeding; Hypothyroidism / Hashimoto's thyroiditis; Hyperthyroidism / Graves disease; Bleeding disorder / Coagulopathy
Interacts with: Propranolol, diltiazem, and many other CYP3A4-metabolized drugs -- guggulsterones induce CYP3A4, potentially reducing drug levels, a broad interaction flag; Anticoagulants -- theoretical additive bleeding risk; Thyroid hormone -- theoretical interaction given guggul's direct thyroid-stimulating activity in animal studies
Already taking it? A rigorous U.S. RCT found no LDL benefit, and possibly even an LDL increase -- worth knowing that before continuing for cholesterol specifically. The thyroid-support claim is not clinically substantiated in humans, only in animal studies.
